2007年5月16日 · Yeah I feel the same way. My old supervisor used to make us prepare them over the flame with sterilized glycerol and then immediately flash freeze them in nitrogen. Personally I find this quite silly, nothing grows at -80. Now I autoclave my 50% glycerol, and mix 250ul of it with 750ul of overnight on my bench and then put it in the -80.

2007年6月8日 · For a 10%wt/vol solution you take 10g of solute mass and suspend it in 1000ml. Or 1g mass in 100ml. Same wt/vol ratio. (In fact, you would suspend the 10g in solvent and make up to 1l..which might not be exactly 1000ml depending on the solute's density).

2010年1月6日 · When preparing high concn agarose it is a good idea to 1) slowly add the agarose powder, with mixing, into the buffer (not the other way around) 2) soak the agarose for 15 minutes at room temperature before heating 3) swirl the flask multiple times during the heating to keep the powder suspended.

I will try to make sure to make a correct dilution (1:1 glycerol-antibody). Ultimately, the staining is what I'm worried about since glycerol could interfere with the visualisation (or not)? And yes, the main reason for doing this is so that antibody stocks doesn't freeze at -20 C since we will be freeze-thawing it regularly.

CXCR2 is easily detected by flow cytometry with FITC or PE conjugated abs. If you don't have access to a flow cytometer, lyse the cells in in 2X gel sample buffer alone by counting your cells and resuspending them in 50µl PBS/1,000,000 cells and add a equal volume of 2XGSB (100mM Tris-Cl pH6.8, 20% Glycerol, 0.2M DTT, 4% SDS, 0.02% bromophenol blue).
